To get a robust error message you need to use a local address. You can get a local address by changing the site binding with IIS Manager. Select the web site in the connections pane, right click and select Edit Bindings. 500.0 - module or isapi error occurred
In the Edit Site Bindings dialog, select the binding (the line in blue highlighting below), then select Edit. Change the IP address from All Unassigned to *. OK, then Close. In my case, the root Web.config of the application had a duplicate section in the the configSections element. The configSections is used for registering all IIS and Windows Activation System (WAS) sections. Config Error : There is a duplicate ‘system.web.extensions/scripting/scriptResourceHandler' section defined I commented out the duplicate line and the application worked. I have been using VS 2010 for developing application pages for SP 2010. I make use of wsp builder to package all my dlls, pages, scripts and images into a solution package and deploy it in the web applications. Adjust your web.config file and set custom errors to off:

The worst thing is .. I am trying to locate the web.config file which i placed at wwwroot, but I couldn't find it... I dont know where it disappeared ,, after creating this mess.. This patch solves the "An authentication error occurred while communicating with the web server. Please see Help for assistance.", and "Debugging failed because integrated Windows authentication is not enabled. Please see Help for assistance" errors, that until now had to be worked around using the instructions below. This patch also enables customers using Windows Vista Home Premium customers to use Visual Studio F5 debugging, who were not able to use this feature at all previously because Windows Authentication was not available on Windows Vista Home Premium.
