If the VPN client bypass TMG server, does it work? If not, you should check the VPN server settings and if there is any other device in front of TMG that block the traffic. Otherwise, please make sure to configure the client behind TMG server as SecureNat, and allow DNS traffic thought TMG server. You may look into TMG logging to see what was happened.
