There is no other process using the file, unless miktex itself uses separate processes to download and extract it and isn't unlocking the file after download, as there is miktex 2.9 download nothing else running. I then close the package manager and re-open the tex file, hit compile and it fails saying it could not install the package. At the end of each updates, the update process end up with the following error message: initexmf.exe: Windows API error 32: The process cannot access the file http://tex.stackexchange.com/questions/274497/miktex-windows-api-error-32 because it is being used by another process. The manual work around is to execute command initexmf --update-fndb --force --mklinks --mkmaps --verbose after each run of update. The problem occurs on Windows XP SP2 (all latest updates) as well as on Vista Home Premium (all latest updates). I first reported this problem on MiKTeX mailing list on 2006-09-11. Since I https://sourceforge.net/p/miktex/bugs/1587/ use simplified Chinese version of these systems, I have no idea whether the problem occurs on systems with other languages, until recently also on the MiKTeX mailing list, Fernando Bastos reported exactly the same problem.
